Desarrollo y aplicación de un método HPLC para la cuantificación de ácido shikímico en frutos de Cotoneaster pannosus: una fuente alternativa de precursores farmacéuticos
Resumen
El ácido shikímico (AS) es el precursor biosintético fundamental para la producción del fármaco antiviral oseltamivir, debido a la necesidad de diversificar sus fuentes naturales, se evaluó el fruto de Cotoneaster pannosus como una alternativa de suministro. Se desarrolló y aplicó un método por HPLC-UV para su cuantificación, empleando una columna ReproSil–Pur Basic-C8, la fase móvil compuesta por agua (HCOOH 1%, pH 2,67) y acetonitrilo en elución por gradiente. La extracción se optimizó mediante ultrasonido a 50 °C, logrando una recuperación eficiente del analito. El método demostró ser lineal (R2: 0,998), exacto (100,1% ± 3,6%) y preciso (CV < 3%), con límites de detección (LOD) y cuantificación (LOQ) de 1,24 ppm y 3,76 ppm, respectivamente. Se determinó una concentración de 50,4 ppm de AS en la muestra fresca. Aunque este contenido es menor al de fuentes tradicionales, la amplia disponibilidad geográfica y la baja complejidad de extracción lo pueden considerar como una fuente natural complementaria de interés para la industria farmacéutica.
